Polly, when you have a minute, could you send ONE specimen of Xylosandrus amputatus collected in Georgia to E. R. Hoebeke at the University of Georgia? We already sent him specimens before.

Adam,
Thank you in advance for your help with Latasha. Here is a summary of what we will do with Latasha on her first visit:

If she has beetles, we will identify them, and grind them up to extract symbionts. Here is how. We will plate them and wait a week.

If she doesn’t have beetles, or only species that are not easy to tell a story about, we will use our fungi.
– Raffaelea subfusca from Craig
– Ambrosiella hartigii from Martin Sigut
– Fusarium sp. from Martin Sigut
– Raffaelea lauricola from Marc
– Diplodia from oaks as a free-living control (or some better species?)

If these fungi are in decently clean cultures, we can subculture them directly into a competitive assay. If they need some cleanup, we will first subculture them alone.

Competitive assay: We will record single-direction reactions: each field has a record of the reactions of the fungus in the upper row. We also need reactions of each to itself, and a growth rate without a competitor. Can we do two replicates of each assay?

How should we score it? How about Strong inhibition, medium, weak, no effect? Or estimated percentage of growth decrease compared to the no-competitor control??

Beetles to Australia

Polly,
Two quick questions: do you ever get any Cleridae (checkered beetles, for example Thanasimus) to your traps? We have an Australian colleague who would send us ambrosia beetles if we sent him some clerids. Please let me know.

Second question: would you please put together several (lets say, four) synoptic collections such as those we sent to Uruguay? There are more colleagues who are asking for such a representative collection. Simply: please put together four sets of approximately 20 to 30 species of beetles that are common around here and of which we have lots. Please let me know if you need more input on this.

– can you make new plates with 2 liters of PDA-hard?
– return the drill please- the sink CAN be used but sparingly. Wash what’s inside.

Polly,
– Where are we on the return of the pipette? Did I send you the Purchase Order?
– Did you happen to deliver the redbay wood to Jane Medley?
– let’s talk about sorting the freezer (please catch me when I’m in)
– we also need to devise a new design for the BTB trapping with new lures. Essentially,w e will do the same thing again with a new Sirex lure (this one is a turpentine from pines rather than from Eucalyptus, like the first one), except the negative control will be replace with a treatment with as many bags of the old stuff as possible (David thinks the old ones may have had low release rate).

Polly, about once a week, could you check for any emerging beetles in rearing boxes in the storage room (the old walk-in fridge)? If there are any, please put them in ethanol and record them in database.
