{"id":1325,"date":"2013-04-10T13:16:48","date_gmt":"2013-04-10T17:16:48","guid":{"rendered":"http:\/\/www.ambrosiasymbiosis.org\/labprotocols\/tube-diets\/"},"modified":"2015-10-19T08:46:05","modified_gmt":"2015-10-19T12:46:05","slug":"tube-diets","status":"publish","type":"post","link":"https:\/\/www.ambrosiasymbiosis.org\/labprotocols\/tube-diets\/","title":{"rendered":"Tube Diets"},"content":{"rendered":"<h1 id=\"firstHeading\"><\/h1>\n<div id=\"bodyContent\">\n<p><b>Diet composition<\/b><\/p>\n<p>Mix:<\/p>\n<ul>\n<li>2.5 g malt extract<\/li>\n<li>44 g sawdust<\/li>\n<li>10 g agar<\/li>\n<li>2.5 g casein (may need to be dissolved in warm water)<\/li>\n<li>5 g dextrose<\/li>\n<li>2.5 g starch<\/li>\n<li>Literature recommends adding antibiotics at this step &#8211; it makes no difference, since you will autoclave this mixture anyway.<\/li>\n<\/ul>\n<p>Add 200 ml tap water and mix thoroughly.<\/p>\n<p>Autoclave mixture and pour in skinny long tubes while hot.<\/p>\n<p>Press with something autoclaved, such as the inner part of a syringe, to squeeze out extra liquid.<\/p>\n<p>After pressing media into tubes and letting solidify, have them freeze and defreeze \u2013 makes media more aerated.<\/p>\n<p><b>Notes<\/b><br \/>\nMalt extract is pretty acidic, so most bacteria don&#8217;t grow there, so antibiotics are not really needed. The bigger problem are molds and mites.
